In general basketball terms, the short answer is yes. Usually the box-and-one is played against an exceptional outside scoring threat and the team's best defender will handle the "and 1" responsibility. If the opposing threat is an interior beast, box and 1 would give way to a more standard 2-3 zone. As for how the game engine will cover it, I don't know. I assume you will decide who the and 1 defender is and maybe -not sure- you will be able to choose who to cover more closely.
I would like to know if playing this defense will accelerate the one vs. one players' fatigue because it sure does in real life.
