I think it is more helpful in NT and U21 games where you have 4-5 players closer in skill level. For traditional club teams, generally the starters you have are substantially better than the backups, so you want your starters playing more minutes. The idea behind blanking 2 spots is that you get a better balance of minutes between all the players and it will boost ratings slightly because the instant one guy is getting tired, there are 2-3 other guys that can sub in for him instead of just one option. Along with this, it gets more combinations of players playing together and the coach can decide the best two players for the 4th quarter and for end game minutes. I think blanking two spots is much more common and effective for U21 and NT.
